壳聚糖合并中药促进肝细胞过氧化物酶体增殖物激活受体α表达的实验研究

【摘要】 目的:研究壳聚糖合并中药对脂肪肝大鼠肝细胞过氧化物酶体增殖物激活受体α(PPARα)表达的影响。方法:在小剂量 CCl4肝损伤的基础上合并高脂饮食建立大鼠脂肪肝模型后,给予壳聚糖合并中药治疗,应用半定量逆转录——聚合酶链反应(RT-PCR)法测定壳聚糖合并中药对大鼠肝 PPARα mRNA表达的影响,并观察大鼠肝组织病理学及肝脏甘油三酯(TG)、总胆固醇(TC)及血清内毒素(ET)、肿瘤坏死因子(TNF)的变化。结果:脂肪肝模型组大鼠肝 PPARα表达明显减少,肝脏 TG、TC水平和血清 ET、TNF含量均明显增加,经药物治疗后肝 PPARα表达基本恢复正常水平,肝 TG、TC水平和血清 ET、TNF含量均明显下降。结论:该复方中药能显著促进脂肪肝大鼠肝细胞PPARα表达,可能是其治疗脂肪肝的分子机理之一。

【Abstract】 Objective To study the effect of chitosan and traditional Chinese medicines on hepatocyte PPARa expression in rat model of fatty liver. Methods The rats were treated with a low dose of carbon tetrachloride (CCl4) and fed a high fat diet to produce fatty liver , then treated with chitosan and traditional Chinese medicines. Changes on pathology of liver and hepatocyte PPARαexpression were observed and the concentrations of endotoxin (ET), tumor necrosis factor(TNF) in serum and TG、TC in liver were detected. Results In model group hepatocyte PPARαexpression decreased significantly and the contents of ET, TNF in serum and TG, TC in liver increased remarkably while in treated group hepatocyte PPARαexpression increased significantly and the concentrations of ET, TNF in serum and TG, TC in liver decreased. Conclusion The chitosan and traditional Chinese medicines could promoting the hepatocyte PPARαexpression markedly in rats with fatty liver, and the effect of treating fatty liver may be associated with this promoting effection.
