亚心形扁藻培养基的筛选与优化

【摘要】 在500mL三角瓶中对亚心形扁藻培养基的筛选与优化结果表明,原“海洋三号”培养基中需添加0.8 g/L的NaHCO3,且 K2HPO4的浓度需由原来的 0.01g/L调整为 0.06 g/L。10 L全自动封闭式光生物反应器分批培养结果表明,优化后的“海洋三号”培养基不仅可加快亚心形扁藻的生长速率而且可延长生长时间,培养 4 d时最高藻细胞密度可达 426×104细胞/mL,比原“海洋三号”培养基的最高培养密度提高42%。优化后的“海洋三号”培养基为亚心形扁藻的最适培养基。

【Abstract】 Compared with other culture mediums, the medium named "marine No. 3" was more suitable for growth of Platymonas subcordifarmis. Further studies indicated that 0. 8 g/ L NaHCO3 should be added to "marine No. 3" medium, and the concentration of K2HPO4 should be changed from 0. 01 g/L to 0. 06 g/L. Results of the batch culture of Platymonas sub-cordiformis in 10 L automatically enclosed photobioreactor showed that the optimized "marine No. 3" medium could not only increase growth rate but also extend growth time of the cell, and the highest cell density reached 426 2 104/ mL after four days, which was 1. 42 times that with the original "marine No. 3"medium.
