重组成骨生长肽对兔同种异体骨移植的成骨影响

【摘要】 目的 观察重组成骨生长肽 (rOGP)静脉应用对兔同种异体骨移植愈合过程中成骨活性的影响。方法 4 2只日本大耳白兔选择一侧前肢造成桡骨中段骨缺损 ,植入常温储存的异体骨 ,随机分成二组。实验组从术后第1d开始每天注射rOGP0 .5 μg kg ,至取材前 1d ,对照组注射同样剂量的生理盐水。术后 4、8、1 2周每组各处死 7只 ,测血清ALP、BGP水平 ,同时取材摄X线片。结果 放射线学显示 ,实验组术后 8周植骨与宿主骨愈合 ,对照组术后 1 2周植骨与宿主骨愈合。实验组血清碱性磷酸酶 (ALP)、骨钙素 (BGP)水平高于对照组 ,术后 4周差异非常显著 (P <0 .0 1 ) ,术后 8、1 2周显著差异 (P <0 .0 5 )。结论 全身应用rOGP通过提高宿主体内成骨细胞的活性 ,促进同种异体骨移植的愈合。

【Abstract】 Objective To investigate the effect of rOGP by intravenous injection on bone formation of healing period of transplantated rabbit bone allograft. Methods Forty two Japanese white rabbits were divided randomly into experimental and control groups,which radius were produced defect and transplantated with stored bone allograft in normal tempreture. The rOGP was administered to experimental animals by intravenous injection 0.5 μg/(kg·d) -1 from first day after surgery to one day before killing, while the control animals were injected with saline solution. Seven rabbits of every group were killed at 4,8,12 weeks after surgery. The serum was obtained to determine the level of ALP and BGP, at the same time the anteroposterior radiographs were taken. Results The bone transplantation site of the experimental animals were healed at 8 weeks after surgery, while the bone transplantation site of the control animals were healed at 12 weeks after surgery on radiograph. The serum ALP, BGP level were higher significantly in rOGP injected group than that in controls at 4 weeks after surgery ( P <0.01),and at 8,12 weeks after surgery,this tendency also could be found( P <0.05).Conclusion Systemically administering rOGP can enhance the healing of allogenic bone transplantation by increasing the osteoblast activity of recipient.
