血糖安对2型糖尿病大鼠小肠α-葡萄糖苷酶mRNA表达的影响
Effect of Xue Tang An on the alvine α-glucosidase’s mRNA expression in the type 2 diabetic rats
【摘要】 目的 观察血糖安对 2型糖尿病大鼠小肠α 葡萄糖苷酶mRNA表达的影响。方法 建立 2型糖尿病大鼠模型。随机分成 5组 :血糖安大剂量组 (9mg kg .d)、血糖安中剂量组 (3mg kg·d)、血糖安小剂量组 (1mg kg·d)、糖尿病模型组、苯乙双胍组 (75mg kg·d)。连续用药 7天用电极试纸法测空腹血糖值及糖耐量值。用小肠α 葡萄糖苷酶原位杂交试剂盒检测小肠α 葡萄糖苷酶mRNA的表达。结果 ① 2型糖尿病大鼠经血糖安给药后 ,空腹血糖值与模型组相比明显降低 (P <0 .0 5 ) ;②用药组大鼠的糖耐量曲线下面积 (AUC)较模型组显著减少 (P <0 .0 1 ) ;③用药组大鼠小肠α 葡萄糖苷酶mRNA的表达与模型组和正常组相比均有明显减少。结论 血糖安对 2型糖尿病大鼠有明显的降糖作用 ,对糖耐量具有保护作用 ,对 2型糖尿病大鼠小肠α 葡萄糖苷酶mRNA的表达有抑制作用
【Abstract】 Objective To study the effect of Xue Tang An(XTA) on the alvine α glucosidase’s mRNA expression in the type 2 diabetic rats.Methods Type 2 diabetes models were established,and were selected into 5 groups randomly:XTA 9mg/kg·d,XTA 3mg/kg·d,XTA 1mg/kg.d,diabetes model group,and phenformin group (75mg/kg·d).The model rats were fed with XTA for 7 days,then assayed the values of plasma glucose and glucosetolerance.The mRNA expression of alvine α glucosidase was assayed by the hybridization in situ technique.Results ①After fed with XTA,the fasting plasma glucose level of the type 2 diabetic rats was obviously lower than that of the diabetes model rats( P <0.05).②The area under glucosetolerance curve was lower in diabetic rats after administration of XTA than that in the diabetes model rats ( P <0.01).③The alvine α glucosidase’s mRNA expression of the diabetic rats fed with XTA is obviously less than that of the normal control rats and the diabetes model rats.Conclusion The study suggested that XTA can decrease the plasma glucose,protect the glucosetolerance,and restrain the mRNA expression of alvine α glucosidase in the type 2 diabetic rats.
