两种新型镍-邻菲咯啉类核酸荧光探针的合成及性质研究

【摘要】 合成了咪唑并[f]邻菲咯啉(IP)和对羟基苯基咪唑并[f]邻菲咯啉(PHPIP)两种配体,并合成了两种新配合物Ni(Phen)2PHPIP和Ni(Phen)2IP,对其进行了表征,用紫外光谱、静态荧光光谱研究了配合物与小牛胸腺DNA的结合情况。两种配合物均与DNA有强烈作用,其中配合物Ni(Phen)2PHPIP在水溶液中几乎无荧光,而加入DNA后,能发射出很强的荧光,可以作为检测核酸分子的荧光探针。

【Abstract】 Two ligands , imidazo [f] 1, 10-phenanthroline (IP) and p-hydroxy- phenylimidazo[f] 1,10-phenanthro-line(PHPIP), and two new complexes , Ni(Phen)2 IP. 2ClO4,Ni(Phen)2 PHPIP.2ClO4 were synthesized and characterized . The binding of the complexes of the complexes to calf thymus DNA was investigated by using steady-state luminescence and ultraviolet visible absorption spectroscopy. The results indicated that the two complexes were bound to CT DNA . Ni(Phen)2 PHPIP showed almost no fluorescence in water , but it produced strong fluorescence when it was bound to CT DNA, so Ni(Phen)2 PHPIP is a prospective new kind of fluorescence probe for nucleic acid determination.
