机采献血者血小板抗原基因分型检测及意义
Genetic Polymorphism and Clinical Significance of Platelet Donors’HPA Genotyping
【摘要】 目的 探讨本地区机采血小板献血者HAP基因多态性分布及意义。方法 用PCR -SSP(序列特异性引物 )方法对 10 0名固定机采献血者进行HPA 1~ 5基因分型检测 ,分析多态性。结果 HPA 1a、4a、5a有较高的基因频率 ,分别为 0 99、0 98、0 96 ,2a、3a基因频率相对较低 ,为 0 88、0 6 4。结论 本地区HPA 1~ 5基因分型结果与韩国、台湾以及国内其他地区一致 ,与欧洲白人存在种族差异。PCR -SSP是一种方便快捷的基因分型方法 ,可进行血小板配型、相关疾病的诊断、遗传多态性调查。
【Abstract】 Objective To investigate the gene frequencies of HPA of the platelet donors and discuss the clinical significance of HPA genotyping Methods One hundred platelet donors were tested for the HPA 1~5 with sequence specific primer polymerase chain reaction (PCR SSP) methods Results HPA 1 a,4 a,5a had the higher gene frequencies of the 5 major HPA alleles (0 99,0 98,0 96),the HPA 2 a,3 a was lower(0 88,0 64) Conclusion The results of the investigation are similar to those reported before.There is significant difference to European studies PCR SSP is a kind of useful method for HPA Genotyping can be used in HPA crossmatch and the diagnosis of alloimmune platelet disease
