中国海南岛黎族3个支系小卫星MSY2多态位点的遗传学研究
POLYMORPHISM ANALYSIS FOR MSY2OF THREE LI POPULATIONS IN HAINAN ISLAND,CHINA
【摘要】 目的:研究中国不同民族Y染色体DNA多态性,追溯人类进化史上的父系祖先,为人类基因组和法医学鉴定积累数据。方法:应用特异引物PCR法扩增Y染色体小卫星DNA片段,再经琼脂糖凝胶电泳检测方法,对海南岛本地黎、杞黎和黎3个支系的293例男性个体的MSY2位点的遗传多样性进行了多态性分析。结果:MSY2各等位基因在黎族3个支系中显示了不同的频率,其中本地黎与杞黎之间无显著性差异,但在本地黎与黎(P﹤0.01)、杞黎与黎(P﹤0.05)之间都存在显著性差异。结论:MSY2位点可以作为一个有用的遗传标记为中国不同群体的遗传多样性研究提供可靠的数据。黎群体可能有复杂的父系起源。
【Abstract】 Objective:The study of Chinese Y chromosome DNA polymorphism is used to trace paternal lineage of human origins and evolution.The data in this study may be useful in human population genetics and forensic applications.Methods:The distribution of a slowly evolution minisatellite on the human Y chromosome ,MSY2,in293unrelated males of three Li ethnic subgroup populations in Hainan island of China was studied by both PCR amplification and agarose gel electrophoresis.The three Li ethnic subgroup populations are Benti-Li,Qi-Li and Ha-Li.Results:The results showed that the MSY23and the MSY24were found at different frequency in three Li subgroups from Hainan Island.There are significant differences between allele frequencies in Benti-Li group and Ha-Li group(P0.01),in Qi-Li group and Ha-Li group(P0.05).Conclusion:These results can be explained by the complex origins of Ha-Li group.As a useful polymorphic marker in Chinese populations,MSY2can provide reliable evidence in evolution study.
