【摘要】 目的 :引进国外先进设备 ,使ABO反定型、ALT、TRUST、HBsAg、抗 HCV、抗 HIV六项检测由手工向自动化操作的转变。提高血站检验的整体检测水平 ,保证临床安全输血。方法 :通过正交试验设计的方法 ,优化最佳试验参数。结果 :TECAN仪器在吸液与分配较粘稠的液体时 ,其系统气隙、拖尾气隙、过量体积需依次设定为 :10、2 0、15 0 μl;TECAN吸ALT丙酮酸标准品的顺序为 ,先吸基质 ,再吸磷酸缓冲液 ,最后吸丙酮酸。不同厂家的微孔板 ,其洗板模式不一样 ,国外的微孔板采用洗板模式为 5 ,国产的微孔板采用洗板模式为 2。血型反定条件为 :转速 10 0 0r/min、离心时间 1min ,振荡悬浮为 90 0r/min ,时间 1min ,振幅为 3。梅毒实验条件为 :转速 15 0 0r/min ,离心时间 2min ,振荡悬浮速度为 75 0r/min ,时间 1min ,振幅为 3 ,静置 3min。条码选择 12 8。局域网联网方式为星型拓扑局域网。结论 :完成了TECAN BEPⅢ全自动筛查仪器在血站系统血液六项检测项目由手工向自动化操作 ,不仅降低了劳动强度 ,而且克服人为因素 ,提高血站检验的整体检测水平 ,保证临床安全输血更多还原

【Abstract】 Objective: Six items,named ABO reverse typing,ALT,TRUST,HBsAg, anti-HCV and anti-HIV,were tested automatically by TECAN-BEPⅢ.These improve testing level of blood station and make clinical transfusion safer.Methods: Orthogonal testing was used to select the best parameter.Results: TECAN instrument system trailing airgap, trailing airgap and excess volume are 10 μl,20 μl and 150 μl when it draws liquid and dispenses thicker liquid. The order of TECAN drawing ALT pyruvic acid standard was first drawing fiducial liquid,then PBS,the last was pyruvic acid.It’s different to wash microplate mode in different factory,the overseas microplate’s mode is five and the internal microplate’s mode is two.The condition of blood reverse typing is centrifugal speed 1000 r/min,1 min,then shaking 900 r/min,1 min,3 accel.The condition of syphilis testing is 1500 r/min,2 min,then shaking 750 r/min,1 min,3 accel,finally placement 3 min.The system’s barcode is 128 code.The local area network is star connection.Conclusion: The authors establish TECAN&BEPⅢ fully automatic screening instrument system to test six items from manual operation to automatic operation,this system not only reduce to work hard,but also overcome manual factors,these improve testing level of blood station and make clinical transfusion safer.更多还原
