河南食管癌高发区居民食管癌和癌旁组织细胞周期素D1蛋白和mRNA的表达

Expression of cyclin D1 protein and mRNA in cancer and adjacent tissues in the same patient in Linzhou, a high incidence area for esophageal cancer in northern China

【摘要】 目的 :探讨细胞周期素蛋白及mRNA表达变化与食管鳞癌发生之间的关系。方法 :采用免疫组织化学过氧化物酶标志的链霉卵白素 (SP)染色法和反转录PCR方法检测食管癌高发区河南省林州市手术标本的细胞周期素D1(cyclinD1)的蛋白表达水平和mRNA表达状况和特征。结果 :癌组织和癌旁组织中均出现不同程度的CyclinD1蛋白和mRNA表达 ,癌旁组织和癌组织CyclinD1免疫组化阳性和mRNA表达阳性率分别为 2 3 1%vs 3 8 4%和 87 5 %vs 81 3 % ,但二者间无显著性差异 (P >0 0 5 ) ,但是 ,碘染色阳性癌旁组织CyclinD1免疫阳性率 ( 2 8 6% )明显高于碘染色阴性组 ( 3 6% )。结论 :CyclinD1与食管鳞癌癌变发生有关 ,可能参与食管癌变的早期阶段

【Abstract】 Objective: To characterize the expression of protein and mRNA of Cyclin D1 in human esophageal carcinogenesis.Methods: Immunohistochemical analysis of Cyclin D1 expression was carried out in paraffin-embedded sections of surgically resected esophageal squmous cancer tissues (n=39) and matched adjacent cancer tissues. Expression of Cyclin D1 mRNA was determined with reverse transcriptase-polymerase chain reactions(RT-PCR) in the same set of samples.Results: Overexpression of protein was observed in the tumour samples and adjacent tissues. The positive protein immunostaining and mRNA expression rates for Cyclin D1 were 23.1% vs 38.4% and 87.5% vs 81.3% in the cancer and adjacent cancer tissues, respectively. But, the difference was not significant(P>0.05). However, the positive immunostaining rate for Cyclin D1 in the samples from positive iodine staining area(28.6%) was much higher than in that from negative iodine staining area(3.6%).Conclusion: The present results showed that overexpression of Cyclin D1 protein and mRNA may participate in esophageal carcinogensis, especially in the early stage of carcinogensis.
