早搏宁冲剂对豚鼠心室乳头肌及窦房结动作电位的影响
Effects of Zaoboning granula on action potentials of guinea pig papillary muscle and sinuatrial node
【摘要】 目的 :观察早搏宁冲剂对豚鼠心室乳头肌及窦房结动作电位的影响。方法 :豚鼠 2 0只分为早搏宁冲剂组 (n =1 0 )与盐酸普鲁卡因胺组 (n =1 0 ) ,2组均采用累积浓度给药法给药 ,常规微电极技术引导豚鼠心室乳头肌快反应动作电位 (APPM)。另取豚鼠 1 6只分为早搏宁冲剂组 (n =1 0 )与盐酸普鲁卡因胺组 (n =6 ) ,同法给药、引导窦房结优势起搏细胞动作电位 (APSN)。示波器照像记录实验结果。观察早搏宁冲剂低、中、高浓度 (0 .1g·L-1 ,0 .2g·L-1 ,0 .3g·L-1 )对APPM及APSN各指标的影响 ,并与盐酸普鲁卡因胺比较。结果 :早搏宁冲剂可降低豚鼠APPM的动作电位幅值 (APA)及 0期最大除极速率 (vmax) ,延长动作电位时程 (APD) ,同时降低APSN的 4期除极速率 (DDR) ,延长窦性周长 (SCL)。其药效弱于普鲁卡因胺 ,但 2药作用性质相似。结论 :早搏宁冲剂具有抗心律失常的作用 ,其机制可能与抑制心肌细胞Na+ 、K+ 通道有关
【Abstract】 Aim: To observe the effects of Zaoboning granula on action potentials of guinea pig papillary muscles and sinuatrial node. Methods: By using microelectrode technique the action potentials on guinea pig papillary muscle (AP PM ) and AP on sinuatrial node (AP SN ) were induced and the effects of Zaoboning granula on AP PM and AP SN were observed. Results: Zaoboning granula at different doses could inhibit the action potential amplitude and maximal upstroke velocity, prolong the action potential duration (APD), ERP (effective refractory period)/APD 90 of AP PM , and suppress the diastole depolarization rate, prolong the sinuatrial circle length of AP SN . Conclusion: The results suggest that Zaoboning granula has the antiarrhythmic effects, which may be embodied by inhibiting Na + and K + channels.
