8-Br-cAMP对Eca-109细胞c-myc,wtp53,iNOS基因和EGFR表达的影响
Differentiation of Eca-109 cell line induced by 8-Br-cAMP
【摘要】 目的 :探讨 8 Br cAMP对Eca 10 9细胞c myc、wtp5 3、iNOS基因和EGFR表达的影响。方法 :将贴壁培养的Eca 10 9细胞随机分为 2组 :①实验组 :加 8 Br cAMP至终浓度为 2× 10 -5mol/L ,培养 2 4h ;②对照组 :不加 8 Br cAMP ,培养 2 4h。对 2组的细胞涂片和硝酸纤维素膜 (NCM)标本进行c myc、wtp5 3、iNOS基因和EGFR表达检测 ,并对扫描数值进行统计学处理。结果 :原位杂交及免疫组化反应信号皆定位于Eca 10 9细胞的胞质 ,扫描数值显示 :①c mycmRNA :实验组为 3.38± 0 .99,对照组为 5 .18± 1.39;②wtp5 3mRNA :实验组为 2 .74± 0 .83,对照组为 0 .38±0 .2 7;③iNOSmRNA :实验组为 4 .5 2± 0 .74 ,对照组为 2 .6 3± 0 .13;④EGFR IR :实验组为 2 .37± 1.0 5 ,对照组为 4 .38±0 .4 8。以上实验组与对照组比较差异有统计学意义 (P均 <0 .0 5 )。结论 :c myc、wtp5 3和NO均可参与 8 Br cAMP诱导癌细胞的分化效应
【Abstract】 Aim: To explore the effects of 8 Br cAMP on differentiation of human esophageal cancer Eca 109 cell line.Methods: The cultured Eca 109 cells were divided into 2 groups: Br group: cells were cultured with 2×10 -5 mol/L 8 Br cAMP for 24 h; Control group: cells were cultured without 8 Br cAMP for 24 h. The in situ hybridization, immunohistochemistry and RNA and protein dot blot technique were used to study the effects of 8 Br cAMP on the expression of genes c myc, wtp53, iNOS, and EGFR IR in each group. Results: In situ hybridization signal and immunohistochemistry IR were all localized in the cytoplasm. The scanning values showed that: a): c myc mRNA: Br group was 3.38±0.99,C group was 5.18±1.39;b): wtp53 mRNA: Br group was 2.74±0.83,C group was 0.38±0.27;c): iNOSmRNA: Br group was 4.52±0.74,C group was 2.63±0.13;d): EGFR IR: Br group was 2.37±1.05,C group was 4.38±0.48; P <0.05~0.01. Conclusion: c myc, wtp53, and NO may be involved in differentiation of Eca 109 cell induced by 8 Br cAMP.
