银屑病患者皮损中细胞衰老、增殖和凋亡检测
Study on the cell senescence, proliferation, and apoptosis in psoriatic lesions
【摘要】 目的 :探讨银屑病患者皮损中细胞增殖、衰老和凋亡的变化。方法 :应用组织化学、免疫组织化学和末端脱氧核苷酸转移酶 (TDT)介导的d UTP 生物素缺口末端标记 (TUNEL)等技术原位检测了 2 0例银屑病患者 (试验组 )皮损和 1 0例正常人 (对照组 )皮肤细胞中与衰老细胞相关的 β 半乳糖苷酶 (SA β Gal)、增殖细胞核抗原 (PCNA)和凋亡细胞。结果 :银屑病皮损中衰老细胞、增殖细胞和凋亡细胞数均比正常人皮肤增多 ,试验组与对照组阳性率分别为 55 % / 1 0 %、80 % / 30 %、70 % / 2 0 % ,经 χ2 检验 ,P <0 .0 5 ,差异有统计学意义。结论 :衰老细胞与凋亡细胞的增加共同对银屑病增殖代谢加快起到部分代偿作用
【Abstract】 Aim: To investigate the cell senescence, proliferation, and apoptosis in psoriatic lesions. Methods: Senescent cell associated β galactosidase(SA β Gal), proliferative cell nuclei antigen(PCNA),and apoptotic cells for 20 cases of psoriasis and 10 normal controls were measured in situ by histochemistry, immunohistochemistry, and terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) techniques. Results: Comparing with normal control, the amount of senescent cells, PCNA positive cells, and apoptotic cells in psoriatic lesions were higher and the positive rates of the senescent cell, proliferative cell, and apoptotic cell in psoriasis group and normal control group were 55%/10%, 80%/30%,and 70%/20%,respectively, and there were significant differences between the 2 groups ( P <0.05).Conclusion: It is suggested that the increase of senescent cells and apoptotic cells in number may be a partial compensation for proliferative metabolism increase in cases of psoriasis.
