Gamma射线照射对体外培养人树突状细胞表型的影响
Gamma irradiation influences phenotype of human dendritic cells in vitro
【摘要】 目的 :了解Gamma射线照射是否影响体外培养的人树突状细胞的表型。方法 :利用含有重组的人GM CSF(80 0U/ml)和IL 4(50 0U/ml)的RPMI 1 640培养基从人的外周血单个核细胞 (PBMC)诱生树突状细胞 (DC)。在培养的第 6d加入 5mg/L的脂多糖 (LPS)继续培养 2 4h促使DC完全成熟 ,于第 7d收获DC并分成 2部分 ,一部分未经Gamma射线照射的DC用作对照组 ,另一部分的DC用 30Gy剂量的Gamma射线照射。采用流式细胞仪分析DC的表面分子。结果 :Gamma射线照射减少树突状细胞CD86 ,CD80和HLA DR ,尤其是CD86分子的表达 (P =0 .0 0 72 )。结论 :Gamma射线照射影响DC的表型
【Abstract】 Aim:To determine whether gamma irradiation influences phenotype of dendritic cells (DCs) in vitro.Methods:DCs were induced from the peripheral blood mononuclear cells (PBMC) with RPMI 1640 medium containing recombinant human GM CSF (rhGM CSF, 800 U/ml) and recombinant human IL 4 (rhIL 4, 500 U/ml). Phenotypic changes were monitored by light microscopy. Lipopolysaccharide (LPS) at a concentration of 5 mg/L was added into the cultures after 6 days of growth and the cells were cultured for another 24 hours for DC complete maturation. The harvested DC on the 7th day were equally divided into 2 parts.One part was used as non irradiated DC (naive DC),and the other part was transferred to a gamma irradiator and irradiated at a dose of 30 Gy (irradiated DC). Cell surface molecules were analyzed by flow cytometry.Results:Gamma irradiation reduced the expression of CD86, CD80,and HLA DR molecules on dendritic cells, especially CD86 molecules ( P =0.007 2).Conclusion:Gamma irradiation of dendritic cells influences phenotype of DC.
