LIF对人胚神经干细胞增殖和分化的影响

【摘要】 目的:探讨LIF对人胚神经干细胞(NSCs)增殖和分化的影响。方法:从孕24周流产胎儿脑组织中分离NSCs,并分4组培养。对照组用基础培养液,第1,2,3实验组分别于基础培养液中加入20μg/L EGF和B27,20μg/L LIF和B27,20μg/L LIF、20μg/L EGF及B27。观察细胞生长情况并在不同时间进行细胞计数。诱导细胞分化后,显微镜下观察。结果:培养3 d细胞开始分裂并聚集成球,后逐渐增大,培养10 d,第1,2,3实验组和对照组神经球形成数分别为(21.5±7.8),(23.8±5.4),(25.2±6.3)和(24.8±6.9),各组间差异无统计学意义。分化实验显示含LIF培养的神经干细胞分化少。结论:在体外,LIF能够促进神经干细胞的增殖并抑制神经干细胞的分化。

【Abstract】 Aim:To investigate the effects of leukemia inhibitory factor (LIF) on (NSCs) the proliferation and differentiation human embryonic neural stem cells. Methods;LIF was added to DMEM/F12-based medium, the growth of NSCs was observed, the number of NSCs was counted at different times, then the cells were induced to differentiate into the main types of central nerve system by altering culture conditions and were viewed under microscope. Results; On the third day, the cells began to divide and formed the neurospheres, then the neurospheres expended increasingly. There were no significant differences between the experimental group and the control group in the number of viable cells and neurospheres. The experiment of differentiation showed that few NSCs differentiated in the culture medium with LIF. Conclusions: LIF can facilitate the proliferation of NSCs and inhibit the differentiation of NSCs in vitro.
