长期直接冻存猪肉样中基因组DNA提取方法的改进与PCR分析

【摘要】 以新鲜猪耳组织作为对照 ,比较了用经典方法从新鲜猪耳组织和长期直接冻存猪肉样中提取的DNA ,提出了从长期直接冻存肉样中提取高质量DNA的改进方法。结果表明 ,采用改进方法可得到大量的、较完整的基因组DNA ,即对于基因组DNA降解较严重的样品是行之有效的方法。对提取到的DNA进行扩增 ,并分析了影响PCR反应的因素。

【Abstract】 The DNA templates,extracted from the fresh tissues of pig ear and from the long term and freezing meat sample of pig by the classical method,were compared the amending method of extracting high quality DNA from long term freezing meat samples was advanced.The result showed that abundant and integrative genome DNA can be extracted from the long term freezing meat sample by the amending method. It is an effective method to those samples in which genome DNA was discomposed severely. The DNA template was amplified further, and the affected factors were also analyzed.
