抗HBsAg单链抗体的等电点测定和结构模拟

【摘要】 应用实验生物学和计算生物学方法对重组人源抗HBsAg单链抗体 (HBscFv)的部分理化性质和空间结构进行分析 .首先应用蛋白质分析软件包Antheprot预测HB scFv等电点 ,然后等电聚焦测定HBscFv等电点 ,在此基础上 ,用神经网络法预测HB scFv二级结构 ,然后用同源建模的方法 ,获得HBscFv三级结构 .结果发现 ,HBscFv等电点理论值为 8.5 1,实验值为 7.3~ 8.1;PHDsec结果表明 ,HBscFv是一种全β蛋白质 ;三级结构建模表明 ,HBscFv的VH 结构域由 9个片层组成 ,VL 结构域由 8个片层组成 ;由于单链抗体的特殊性 ,三级结构建模无法给出VH 与VL 结构域之间进一步折叠后形成的结构 .

【Abstract】 To identify the 3D structure and physical and chemical character of a single-chain Fv against HBsAg (HBscFv),methods of experimental biology and computational biology were applied. First,Antheprot,a comprehensive software to analyze protein,was used to predict the isoelectric point (pI) of HBscFv. Second,the predicted result was confirmed by IEF. Third,neural network systems (PHDsec) and SWISS-MODEL were applied to predict the secondary and tertiary structure of HBscFv,respectively. It was found that the theoretical pI of HBscFv is 8.51 and the experimental pI is from 7.3 to 8.1. PHDsec indicated that HBscFv is a whole β -globin. The predicted tertiary structure displayed that the V H domain of HBscFv was made up of 9 β -sheets and V L domain of 8 β -sheets. Due to the specific characteristics of single-chain Fv,SWISS-MODEL software for homo- logy modeling of tertiary structure cannot predict the quaternary structure resulting from the interaction of V H and V L domains.
