二氧化硫对大鼠肺泡巨噬细胞DNA的损伤作用
Damage effects of sulfur dioxide inhalation on DNA damages in alveolar macrophages from male rats
【摘要】 运用单细胞凝胶电泳技术 (又称彗星实验 )研究了SO2 动式吸入对雄性Wistar大鼠肺泡巨噬细胞DNA的损伤作用 .结果表明 ,在SO2 浓度为 2 8 6,57 3 ,114 4mg m3 的动式吸入染毒条件下 ,雄性大鼠肺泡巨噬细胞DNA拖尾长度分别为 7 59,12 3 9,2 1 2 2 μm ,表明SO2 可引起大鼠肺泡巨噬细胞DNA损伤 ,且随SO2 吸入浓度的增加而加剧 ,呈明确的剂量效应关系 .这意味着SO2 具有引起哺乳动物肺泡巨噬细胞DNA突变的潜在危险 ,会造成机体的非特异性吞噬功能和抗肿瘤免疫监视作用的损伤
【Abstract】 The effects of SO2 dynamic inhalation on DNA damages in alveolar macrophages of male rats were studied with the single cell electrophoresis technique(Comet assay).The results showed that after exposure to SO2 at 28.6,57.3,114.4mg/m+3,tail lengths of nuclear DNA in the alveolar macrophages from male rats were 7.59,12.39,21.22μm,respectively.SO2 could lead to DNA damages in alveolar macrophages in a dose-dependent manner.It implies that SO2 has a potential risk to cause DNA mutation of alveolar macrophages from mammalian animals,and it may cause damages to the engulf function and anti-tumor function of alveolar macrophages.=
