CRP和LPS致正常人外周血单核细胞NF-κB活化及IL-6合成的比较
Comparison of Interleukin-6 Synthesis and NF-κB Activation in Monocytes Stimulated by C-reactive protein and Lipopolysaccharide
【摘要】 目的 :观察C -反应蛋白 (CRP)和脂多糖 (LPS)诱导人外周血单核细胞合成白细胞介素 - 6水平及两者致单核细胞NF -κB活化的程度 ,比较其致炎作用。方法 :Ficoll密度梯度离心法分离人外周血单核细胞 ,应用酶联免疫吸附试验 (ELISA)法观察CRP和LPS刺激单核细胞产生IL - 6的时间效应 ,比较其峰值。免疫细胞化学观察两者致单核细胞NF -κB活化的程度。结果 :CRP和LPS刺激IL - 6合成开始的时间分别是 4小时和 2小时 ,呈时间依赖性 ,在 2 4小时达高峰。LPS组峰值高于CRP组。在刺激 1 6小时后 ,LPS组NF -κB活化的程度高于CRP组。结论 :CRP和LPS均能活化NF -κB ,并诱导单核细胞产生IL - 6 ;1 0ng/mlLPS的致炎作用强于 2 0ug/mlCRP。
【Abstract】 Obijective:To compare proinflammatory action of c-reactive protein(CRP) and lipopolysaccharide by measuring interleukin-6(IL-6) production and nuclear factor kappa B(NF-κB)activation in human monocytes.Methods:Monocytes were isolated by Ficoll density gradient from blood of healthy volunteers.To measure IL-6 production (by ELISA)stimulated by 20 ug/ml CRP and 10ng/ml LPS for 2h,4h,8h,16h,24h,the peaks to be compared with each other.NF-κB activation of monocytes were evaluated by immunocytochemisty.Results:IL-6 could by produced after being stimulated with CRP at 4h and with LPS at 2h.The effects of CRP and LPS were time-dependent,reaching peaks at 24h.The peak of IL-6 in LPS group was higher as compared with CPR group.NF-κB activation rate was lower in CRP group after being stimulated for 16h.Conclusions:CRP and LPS can activate NF-κB and induce the production of IL-6 in monocytes;the proinflammatory action of 10 ng/ml LPS is stronger thean that of 20ug/ml CRP.
