胆碱酯酶抑制法检测辛硫磷农药残留
A Method to Detect Phoxim Residue by Analyzing the Inhibition of Cholinesterase
【摘要】 利用胆碱酯酶水解碘化硫代丁酰胆碱后形成的硫代胆碱 ,与二硫双对硝基苯甲酸发生化学反应生成黄色产物 ,在 412nm比色测定 .利用此法测定酶、底物的添加量以及保温温度和保温时间对酶活力的影响 ,还研究了不同浓度与保温时间的辛硫磷对胆碱酯酶活力的抑制情况 .获得了胆碱酯酶优化的反应条件 ,结果还表明胆碱酯酶对辛硫磷比较敏感 ,其抑制中浓度为I50 为 3 .90 49μg/L .
【Abstract】 Ellman reaction is a simple and sensitive method to analyze cholinesterase activity. A new found cholinesterase was studied to get the best reaction condition through Ellman reaction, and it was found this cholinesterase was very sensitive to phoxim, its medium inhibition concentration I 50 to this cholinesterase was only 3.904?9?mg/L, so it had good promise to be used in detecting organophosphate and carbamate pesticide residue in agricultural products.
