国产柴油机废气颗粒提取物致NIH3T3细胞转化作用的实验研究
Study on Transformation Effect of Diesel ExhaustParticulate on NIH3T3 Cell
【摘要】 目的 评价 3种国产柴油机 (6 12 0型、 4 115型、S - 195型 )废气颗粒提取物致细胞转化活性 ,预测它们的致癌性。方法 以国产 3种柴油机废气颗粒提取物为受试物 ,采用细胞转化技术 ,观察其致NIH3T3细胞形态学转化作用。结果 国产 3种类型柴油机废气颗粒提取物均可致NIH3T3细胞形态转化 ,且转化率存在剂量 -反应关系 ,随着剂量的加大 ,细胞转化率增高 ,除 6 12 0型和 4 115型的 0 2 5 μg/ml 剂量组外 ,其他受试物不同剂量组的细胞转化率与对照组比较 ,差异均有显著性 (P <0 0 5 )。相同剂量下S - 195型柴油机废气颗粒提取物的细胞转化活性较 4 115型和 6 12 0型高。结论 柴油机废气颗粒提取物具有不同程度的致细胞转化作用 ,柴油机废气颗粒物的职业性危害不容忽视。
【Abstract】 Objective To evaluale cell transformation activity of exhaust particulates from three kinds of domestic made diesel engines on NIH3T3 cells line and their carcinogenictity.Methods Morphological transformation of NIH3T3 cells induced by three kinds of diesel exhaust particulates were observed using cell transformation technique.Results All these three kinds of diesel exhaust particulates could lead to morphological transformation of NIH3T3 cells and transformation rates increased with the dose.Compared with the contol group,transformation rates were significantly different(P<0 05)for different dose group except for types of 6120 and 4115 diesel engines at 0 25 μg/ml .By giving the same dose,the transformation activity of exhaust particulate from S-195 diesel engine was higher than those from 4115 and 6120 diesel engines.Conclusions Exhaust particulates from diesel engines can lead to transformation of NIT3T3 cells and this occupational hazard should not be neglected.
