静态负荷家兔血清肌型肌酸激酶同功酶和乳酸脱氢酶同功酶的变化
Changes of Serum Creatine Kinase Isoforms and Lactate Dehydrogenase Isoforms in Rabbits with Static Load
【摘要】 目的 观察家兔在静态负荷过程中血清肌酸激酶 (CK)、肌型肌酸激酶同功酶 (CK MM )和乳酸脱氢酶 (LDH)同功酶活力的变化。方法 将实验用家兔分为 2组 ,观察组家兔腰部施加一定的静态负荷 ,另一组为对照组 ,分别于实验的不同时间取血 ,应用琼脂糖凝胶电泳法、聚丙烯酰胺凝胶电泳法和生化法测定两组动物血清中总CK及CK MM、总LDH及其同功酶的活力。结果 观察组动物血清CK活力都随实验时间的延长而升高 ,各时间点的血清CK活力与对照组比较 ,差异有显著性 (P <0 0 1) ;观察组的LDH活力随时间延长从 3h开始与对照组比较 ,差异有显著性 (P <0 0 5 ) :观察组CK MM3 在实验的 3h开始明显升高 ,CK MM3 /CK MM1比值也明显升高 ,与对照组比较 ,差异有显著性 (P <0 0 1)。观察组LDH同功酶的活力变化显示 ,从 3h开始 ,LDH5升高明显 ,LDH高于对照组 (P <0 0 5 ) ,6h达到最大值。结论 静态负荷致肌肉损伤过程中 ,血清中的CK总活力改变是较早出现的敏感指标。CK MM3 /CK MM1发生明显变化 ,LDH的肌型同功酶LDH5也显著升高 ,反映出肌肉组织有新的损伤出现
【Abstract】 Objective To observe activity changes of serum creatine kinase(CK),creatine kinase isoenzyme(CK\|MM) and lactate dehydrogenase(LDH) isoforms in rabbits with static load.Methods The experimental rabbits were divided into experimental group and control group.The experimental group was exerted with some static load on their lumbar.Blood samples were collected from these groups at different time period.Agarose gel electrophoresis method,SDS PAGE method and biochemical method were used to measure activities of serum CK;LDH,and their isoforms.Results It was found that the activity of serum CK increased with the time.Compared with the control group,activity of serum CK showed significant difference at different time period. (P<0 01). LDH activity and its isoform,LDH 5,in the experimental group increased with the time.Begining from the 3 rd hour,they showed significant difference with the control group (P<0 05). 6 hours later,LDH 5 reached the highest.CK\|MM 3 also elevated 3 hours later and for both CK MM 3 and the ratio of CK\|MM 3/CK\|MM 1,significant difference existed between the experimental group and the control group (P<0 01). Conclusions Results indicated that during muscle injury process induced by static load,activity changes of total CK and LDH in serum might be the early and sensitive indicators.The elevated CK\|MM 3/CK\|MM 1 and LDH 5 might reflect muscle tissues injury occurred in recent time.
