发酵液L-亮氨酸的定量测定方法研究——纸色谱-薄层扫描测定法

【摘要】 将纸色谱和薄层扫描相结合,建立一种比较简便、快速、准确地定量测定发酵液L-亮氨酸含量的新方法。发酵液预处理后,用纸色谱分离、茚三酮显色,得到L-亮氨酸色斑;用岛津CS-930薄层扫描仪测定L-亮氨酸标样和发酵液样品的色斑峰面积后,计算出发酵液L-亮氨酸含量。结果表明,本方法的重现性和准确性较高,适用于大批量测定发酵液中的L-亮氨酸含量。

【Abstract】 The combination of paper chromatography and thinlayer scanning may set up a new method of easily, quickly and correctly determining the amount of Lleucine in fermented liquid After being pretreated, fermented liquid is separated by paper chromatography and the colour is shown by ninhydrin, the Lleucine mottle is obtained Using the thinlayer scanner of shimadzu CS-930 to determine the samples of Lleucine and fermented liquid, the amount of Lleucine in fermented liquid are calculated The results show that the method is of higher recurrence and accuracy and may be used to massively determine the amount of Lleucine in fermented liquid
