农杆菌介导BADH基因转化葡萄的研究

【摘要】 以葡萄品种鲁贝的花丝为试材,在葡萄的细胞悬浮系得到胚性愈伤组织及悬浮细胞再生植株的基础上,通过农杆菌介导转化甜菜碱醛脱氢酶基因,利用共培养后GUS染色组织分析探讨了影响转化效率的各种因素,优化了转化体系。组培苗经PCR检测,证明获得了转基因葡萄植株。

【Abstract】 This experiment had been conducted from1999to2001.The flower filaments of some grape varieties were used as materials.On the basis of embryotic callus obtained from grape cell suspension and the plantlets generated from cell suspen-sion system,the BADH gene was transformed through Agrobacterium-mediated way.Factors affecting the transformation fre-quency were studied and optimized transforming system by using GUS coloration after co-culture.The plantlets obtained were analyzed by PCR and were proved that the BADH gene had integrated into the grapevine genome.
