高浓度糖对体外培养的视网膜Müller细胞膜离子通道的影响(英文)

【摘要】 目的观察在高浓度葡萄糖条件下,体外培养的兔视网膜Müller细胞的钙依赖性钾通道的变化。方法高浓度葡萄糖条件下体外培养兔视网膜Müller细胞,应用免疫组织化学技术和透射电镜鉴定Müller细胞,并采用膜片钳技术观察不同时期高糖条件下视网膜Müller细胞钙依赖性钾通道的变化。结果高糖对体外培养的Müller细胞钙依赖性钾通道有阻滞作用,其阻滞作用呈时间依赖性。结论高浓度葡萄糖可能通过阻滞钙依赖性钾通道而降低Müller细胞的生物学功能。

【Abstract】 Aim To observe the effects of high concentration glucose on the calci um-dependent-potassium channel of rabbits’ retinal Müller cells. Methods The rabbits’ retinal Müller cells were cultured in vitro under the condition of hi gh concentration glucose, and identified by immunohistochemical staining and tra nsmission electron microscopy. Patch-clamp technique was used to observe the cha nge in the calcium-dependent-potassium channel in retinal Müller cells caused b y high concentration glucose at different periods. Results High concentration g lucose could inhibit the calcium-dependent-potassium channel of cultured retinal Müller cells in a time-dependent manner. Conclusion High concentration glucos e may reduce the biological functions of Müller cells by inhibiting calcium-dep endent-potassium channel.
