无血清培养的牛体外受精胚胎超快速玻璃化冷冻
Vitrification of Bovine IVF Embryo Produced in Serum Free Medium
【摘要】 采用BSA替代体外发育培养系统SOFaaECS中的怀孕母牛血清来培养牛的体外受精早期胚胎,并且利用OPS超快速玻璃化冷冻技术对处于囊胚期的胚胎加以冷冻。研究结果表明,BSA在SOF aa培养体系当中完全可以替代血清的作用,并且取得了稳定可靠培养的效果,囊胚发育可以达到47%,胚胎的生产效率在稳定基础上得到提高。无血清培养体系生产的OPS玻璃化冷冻胚胎解冻后经培养,存活率达87.8%。利用这一培养系统和冷冻技术能够降低胚胎生产的成本,简洁有效,提高体外受精技术的实用性,为这一技术的推广提供了有益的参考。
【Abstract】 The bovine in vitro fertilized embryo were cultured in the serum free medium SOFaaBSA, to investigate the consequence of BSA instead of ECS, which used to be put in to SOFaa medium .The embryos were freezed by the OPS-a vitrification method, then thawed and cultured to exam the further development potential . The result showed BSA could successfully substitute ECS in the culture medium, blastocyst rate in SOFaaBSA was 47% , compare with the control group 37.3%. After thawing and 24hrs culture, 87.8% embryos produced from serum free medium were re-expanded. It indicated that SOFaaBSA medium be a reliable culture system with more defined components, and make in vitro production of bovine embryos stable.
