海南省恶性疟原虫MSP1和MSP2等位基因分型研究
Study on the genotypes of MSP1 and MSP2 genes of Plasmodium falciparum isolates from Hainan Province
【摘要】 目的 对海南省恶性疟原虫分离株的裂殖子表面蛋白质 1(MSP1)和裂殖子表面蛋白质2 (MSP2 )基因进行分型研究。方法 从海南省恶性疟流行区采集的恶性疟患者血样 ,用套式PCR方法分别扩增PfMSP1和PfMSP2基因中具有型特异性的片段 ,进行等位基因分型。结果 (1)MSP1基因分型 :94份恶性疟患者血样中有 82份扩增出MAD2 0型基因片段 (占 87 2 % ) ,2 7份扩增出K1型基因片段 (占 2 8 7% ) ,15份同时扩增出MAD2 0型和K1型基因片段 (占 16 0 % ) ,未扩增出RO33型基因片段。 (2 )MSP2基因分型 :94份血样中有 75份扩增得到 3D7型基因片段 (79 8% ) ,2 8份扩增得到FC2 7型基因片段 (2 9 8% ) ,10份同时扩增得到 3D7型和FC2 7型基因片段 (占 10 6 % )。结论 海南省恶性疟原虫的MSP1等位基因和MSP2等位基因分别以MAD2 0型和 3D7型为优势基因型 ;不同等位基因型的混合感染率很低
【Abstract】 Objective To type the Merozotite Surface Protein 1 (MSP1) and Merozotite Surface Protein 2(MSP2) genes of Plasmodium falciparum isolated from Hainan Province.Methods Nested PCR technique was applied in the genotyping of MSP1 and MSP2 genes of P. falciparum isolated from blood samples of malaria patients. Results (1) MSP1 genotypes: 94 blood samples, MAD20 type and K1 type alleles were obtained in 82 (87.2%), 27 (28.7%) samples respectively and both alleles in 15 (16 0%) samples. No RO33 type allele was found. (2) MSP2 genotypes: 94 blood samples, 3D7 type and FC27 type alleles were obtained in 75(79.8%),28(29.8%) samples respectively and both alleles in 10(10.6%) samples.Conclusion MAD20 type MSP1 allele and 3D7 type MSP2 allele were dominant in P. falciparum population in Hainan Province. The mixed infection rate of different types of MSP1 or MSP2 alleles was low.
