不同Waxy基因型水稻的遗传多样性
Genetic Diversity of Different Waxy Genotypes in Rice
【摘要】 利用均匀分布于水稻基因组的 2 38个微卫星标记对包含 15种Waxy复等位基因的 4 1个水稻品种(系 )的遗传多样性进行了分析。结果表明有 2 2 1对SSR引物在供试材料间检出多态性 ,共检出 115 6个等位基因 ,平均每个座位可检出 5 2 3个等位基因 ;各个座位的多态信息量 (Polymorphisminformationcontent ,PIC值 )的变幅为 0 0 5 - 0 89,平均PIC值为 0 5 9。聚类分析结果表明 :4 1个供试品种 (系 )明显地聚为两大类群 ,分别对应于籼亚种和粳亚种 ,不同 (CT)重复次数的品种 (系 )在两大类群中有明显的分布规律 :(CT)重复次数较少 (n =8、 10、 11和 12 )的品种 (系 )只出现在籼稻类群中 ,粳稻类群的重复次数相对较多 (n =14 - 2 1) ,而 (CT) 17和 (CT) 18的品种 (系 )中既有属于籼稻类群的 ,又有属于粳稻类群的 ;Waxy基因第一内含子 5′端剪切位点上的G -T单核苷酸多态与籼粳分类没有直接联系。 4 1个品种 (系 )之间的遗传相似系数介于 0 13- 0 88之间 ,平均相似系数为 0 35。Waxy座位上丰富的等位基因 ,以及品种间较高的遗传多样性 ,为水稻的品质育种提供了丰富的种质资源
【Abstract】 Genetic diversity of 41 rice cultivars containing 15 multi alleles at the waxy locus, were investigated using two hundreds and thirty-eight SSR markers which distributed evenly on 12 rice chromosomes. Two hundreds and twenty-one polymorphic markers detected 1156 alleles, with an average of 5 23 alleles per SSR locus. The PIC (Polymorphism information content) value of the polymorphic markers varied from 0 05 to 0 89, with an average of 0 59. Through cluster analysis based on the 238 SSR markers, the 41 cultivars could be classified into two distinct groups, which corresponded to the indica and japonica subspecies, respectively. At the waxy locus , lower (CT)n repeats(n=8,10,11,12) were found only in indica varieties while japonica varieties contained higher (CT)n repeats (n=14-21), but (CT) 17 and (CT) 18 alleles occurred both in indica and japonica varieties. G-T polymorphism at the Waxy leader intron 5′splice site was not correlated with the classification. Genetic similarity coefficient among 41 cultivars ranged from 0 13 to 0 88. The multiple alleles at the waxy locus and the genetic diversity in the varieties provide a base for grain quality improvement in rice.
