紫菜多糖的提取及抗流感病毒活性研究

【摘要】 采用4种方法提取坛紫菜多糖,其中综合法提取率高达16.58%.气相色谱及化学分析表明其单糖主要由半乳糖、3,6-内醚-L-半乳糖、甘露糖和少量木糖组成.可溶性多糖经DEAE-23柱层析进一步纯化,得到蒸馏水洗脱组分———中性多糖和1.0-2.0mol LNaCl梯度洗脱组分———酸性多糖,各占4.12%和95.88%.酸性多糖经硫酸降解和超滤膜分离得到多糖组分F1(3500<MW<30000)和F2(MW>30000),二者比例为4∶1.鸡胚实验表明,紫菜多糖具有抗甲1型流感病毒活性,且F1>F2>中性多糖.

【Abstract】 Porphyra haitanensis polysaccharide was extracted by four methods, among those that the extracting rate of synthesis method was the highest, reaching 16.58%. As shown by the gas chromatographic and chemical analysis, the monosaccharide of soluble polysaccharide is comprised of galactose, 3, 6-AG, mannose and a little xylose. The crude polysaccharide was purified by DEAE-23 and the neutral polysaccharide was eluted by distilled water and the polyanion polysaccharide gradually by 1.0-2.0?mol/L NaCl. Their contents were 4.12% and 95.88%, respectively. The polyanion polysaccharide was degraded by vitriol and separated by ultrafiltration .The polysaccharide F1(3?500<MW<30?000) and F2(MW>30?000)were obtained, the proportion of 4∶1. The chick embryo experiments showed that the porphyra polysaccharides had antiinfluenza A1 virus activity , and F1>F2> neutral polysaccharide.
