膜抗原单抗对痢疾杆菌入侵HeLa细胞细胞骨架的影响

【摘要】 目的:观察抗痢疾杆菌膜蛋白IpaB和LPS的单抗对痢疾杆菌入侵HeLa细胞细胞骨架的影响。方法:采用不同膜抗原的单抗处理痢疾杆菌后进行HeLa细胞入侵及阻断实验,观察不同膜抗原单抗对痢疾杆菌入侵的作用。结果:观察到抗菌膜蛋白IpaB的单抗和LPS的单抗均不能阻断细菌的入侵且能促进细菌的入侵,单抗作用后受感染的HeLa细胞胞内肌动蛋白极性分布现象更为显著,入侵的细菌数量明显增多。结论:提示痢疾杆菌的膜蛋白和LPS单抗所识别的表位,均具有使细胞骨架发生改变、调节S.flexneri2a 12菌进入HeLa细胞的能力。

【Abstract】 Objective:To study the effect of McAbs against S.flexneri2a12 membrane protein and LPS on the bacteria’s invasion to the cytoskeleton of Hela cell. Methods:After S.flexneri2a12 treated with McAbs against various membrane antigens, S.flexneri2a12 invaded the HeLa cells. The invasion results were analyzed by laser scanning confocal microscopy. Results:Neither McAbs against S.flexneri2a12 membrane protein IpaB or LPS could not prevent S.flexneri2a12 invasion. On the contrary,both of them could promote the invasion process. In fact, after treatment with these McAbs, actin’s polar distribution in infected HeLa cells became more obvious. Conclusion:It suggests that McAbs against membrane protein and LPS recognize such epitopes that are able to regulate cytoskeleton to facilitate the entrance of S.flexneri2a12 into Hela cells.
