【摘要】 目的 :探讨hTERT和c mycmRNA在膀胱移行细胞癌 (TCC)患者尿脱落细胞和外周血细胞的表达、临床意义及关系 .方法 :采用RT PCR法同时检测膀胱肿瘤细胞株 ,2 8例TCC患者和 15例正常对照尿脱落细胞和外周血的hTERT和c mycmRNA表达 .结果 :尿脱落细胞hTERT和c myc阳性表达率分别为 85 .7%和 71.4 % ,对照组为 13.3%和 2 6 .7% ;血细胞两指标的阳性表达率分别为 78.6 %和 5 7.1% ,对照组为 6 .7%和 33.3% ;肿瘤组与对照组比较具有显著性差异 (P <0 .0 1) ,两指标不同组间比较具有相关性 (P <0 .0 1) .结论 :hTERT和c mycmRNA在TCC中的表达具有相关性 ;二者可作为膀胱癌的良好诊断指标 ,hTERT较c myc特异和灵敏 ;尿脱落细胞较血细胞对TCC的诊断更准确和方便更多还原

【Abstract】 AIM: To investigate the expression, correlation and clinical values of human telomerase reverse transcriptase (hTERT) and c myc mRNA of urine exfoliative and blood cells in transitional cell carcinoma of the bladder (TCC). METHODS: Using reverse transcription polymerase chain reaction (RT PCR) assay, we simultaneously examined hTERT and c myc mRNA in bladder cancer T24 cell line, 28 TCC patients’ blood samples, urine exfoliative cells and 15 control samples. RESULTS: hTERT and c myc mRNA were detected in 24 of 28 (85.7%) and 20 of 28 (71.4%) respectively in TCC urine samples, while 2 of 15 (13.3%) and 4 of 15 (26.7%) in controls. In blood samples, the two items were detected 22 of 28 (78.6%) and 16 of 28 (57.1%), while 1 of 15 (6.7%) and 5 of 15 (33.3%) in controls. There was significant difference between the positive rate of hTERT and c myc in tumor group and controls ( P < 0.01), and a correlation was found between the expression of hTERT and c myc mRNA in tumor group. CONCLUSION: The expression of hTERT and c myc mRNA is related in TCC, especially in urine exfoliative cells. Both hTERT c myc can be used as diagnostic markers but the former is more sensitive and specific than the latter.更多还原
