抑癌基因PTEN对人前列腺癌细胞系PC-3的作用

【摘要】 目的 :研究抑癌基因PTEN对人前列腺癌细胞系PC 3细胞生长、粘附、细胞周期、超微结构的影响 .方法 :利用阳离子脂质体LipofectAmine2 0 0 0介导 ,分别将含PTEN的逆转录病毒载体PBabe PTEN puro及空载体PBabe puro转入PC 3细胞 ,观察转染前后细胞的生长、粘附、超微结构、细胞周期改变情况 .结果 :将PTEN转入后 ,细胞生长明显受到抑制 ,生长抑制率为 83 %;同质型粘附率下降 ;细胞超微结构失去部分恶性表型特征 ;细胞周期由G1→S0期受抑制 ,并且出现一个小的凋亡峰 .结论 :PTEN可以明显的抑制PC 3生长、粘附 .因而PTEN可能会在前列腺癌基因治疗上起一定的作用

【Abstract】 AIM:To study the effects of tumor suppressor gene PTEN on human prostate cancer cell line PC 3. METHODS: Cationic liposome LipofectAmine2000 was used to transfect retrovirus vector Pbabe PTEN puro containing PTEN and Pbabe puro into PTEN deficient prostate cancer cell line PC 3 respectively, and then cell growth, adhesion, segregation, apoptosis, cell cycle and ultrastructure alteration in vitro were studied. RESULTS: After PTEN was transfected into PC 3 and expressed, cell growth rate obviously slowed down and the growth inhibition rate was 83%. Adhesion rate dropped and cell segregated from matrix more easily. FCM showed a small apoptosis peak and an obvious G1 block in cell cycle and EMS showed that cells lost some malignant features in ultrastructure. CONCLUSION: PTEN can inhibit PC 3 growth and adhesion in vitro and may play an important role in prostate cancer gene therapy.
