藏药独一味两种方法提取组分对小鼠镇痛作用的影响
Analgesic effects of different extraction components from Lamiophlomis rotata (Benth.) kudo, a medicinal plant in XiZang (Tibet) in mice
【摘要】 目的 :中药化学提取分离技术与药理学试验方法相结合 ,构建中药有效成分研究的实验方法 .方法 :以独一味为代表药物 ,采用溶剂萃取法及大孔吸附树脂梯度洗脱分离法两种方法制备样品 ,通过小鼠醋酸扭体法及热板法筛选镇痛有效部位 .结果 :两种方法提取分离的样品对醋酸引起的小鼠扭体反应有明显的抑制作用 ;大孔吸附树脂梯度洗脱法分离的样品能明显延长小鼠热板的痛阈时间 .萃取法中镇痛有效物质主要集中在正丁醇部分、石油醚部分、水部分 ;大孔吸附树脂梯度洗脱分离法中主要集中在 30 0mL·L-1乙醇洗脱部分、90 0mL·L-1乙醇洗脱部分和沉淀物部分 .结论 :初步建立了中药有效部位的研究方法 ,找到了独一味中镇痛有效部位
【Abstract】 AIM: To explore an experimental method for the study of effective components in traditional Chinese medicine by chemical separation technology integrated with pharmacological experimentation. METHODS: Lamiophlomis rotata , a medicinal plant in Tibet, was chosen in our study and different compounds were separated by two methods: extraction with solvent and gradient elution with macroporous adsorptive resin. Effective components of analgesic were screened by acetic acid writhing and hot plate test in mice. RESULTS: Samples separated by the two methods had obvious analgesic action on writhing reaction by acetic acid in mice and could prolong the latencies of paw licking to gradient elution with macroporous adsorptive resin. N butanol extract, petroleum ether extract and water extract were the main effective components of analgesic in extraction with solvent. 300 mL·L -1 ethanol, 900 mL·L -1 ethanol elution parts and precipitate were the effective components of gradient elution with macroporous adsorptive resin. CONCLUSION: Experimentation on effective components of traditional Chinese medicine has been conducted and effective analgesic components are found in Lamiophlomis rotata (Benth.) kudo.
