大鼠脊髓压迫损伤减压后钙离子和兴奋性氨基酸的变化

【摘要】 目的 :SS研究大鼠脊髓压迫损伤减压后钙离子和兴奋性氨基酸变化及其与神经功能恢复的关系 .方法 :将动物随机分为 :慢性渐进性脊髓损伤组 ,慢性渐进性脊髓损伤减压 1,7,14 ,2 8d组 .取伤段脊髓 ,用高效液相色谱法检测谷氨酸和天门冬氨酸的含量的变化 ,用原子吸收光谱法测定其离子改变及水含量 .结果 :慢性渐进性脊髓损伤减压后钙离子和兴奋性氨基酸有明显的降低 ,减压后 1,7,14 ,2 8dGlu(μmol·g-1)分别为 :6 .16± 0 .16 ,5 .4 3± 0 .2 2 ,5 .4 3± 0 .17,5 .33± 0 .2 0 ;Asp(μmol·g-1)分别为 :3.39± 0 .12 ,2 .96±0 .11,2 .92± 0 .16 ,2 .89± 0 .15 ;Ca2 + 离子 (μmol·kg-1)分别为 :5 .84± 0 .11,5 .33± 0 .10 ,5 .2 8± 0 .17,5 .2 4± 0 .0 2 .它们均较减压前 (Glu为 6 .32± 0 .10 μmol·g-1,Asp为 3.5 6±0 .13μmol·g-1,Ca2 + 为 6 .5 3± 0 .6 4 μmol·kg-1)有显著降低 (P <0 0 5 ) ,其中前 7d降低较快 ,以后有降低不明显 .减压后 1,7,,14 ,2 8d改良的Tralov评分分别为 3.5 3± 0 .11,3.74± 0 .12 ,3.86± 0 .12 ,3.88± 0 .10 ;斜板试验分别为 4 3.3± 2 .3,4 6 .2± 1.1,4 8.8± 2 .8,4 9.7± 2 .6 ,均较减压前Tralov评分 (3.4 0± 0 .13)斜板试验 (40 .9± 1.8)有显著升高

【Abstract】 AIM: To study the changing contents of excitatory amino acids and calcium ions and their relation to neuronal function recovery after decompression of spinal cord compression injury in rats.METHODS: Experimental rats were divided into eight groups at random:chronic gradual spinal cord injury group; chronic gradual spinal cord injury decompression 1, 7, 14, 28 d group. Reginal concentrations of excitatory amino acids of the impaired spinal cord were assessed with high performance liquid chromatography. Regional water and ion contents of the impaired rat spinal cord tissue were measured with the atomic absorption spectrophotometry. RESULTS: The contents of excitatory amino acids and calcium ions after chronic gradual spinal cord injury decompression in rats at 1, 7, 14, 20, 28 d (the Glu(μmol·g -1 )were :6.16±0.16 , 5.43±0.22, 5.43± 0.17, 5.33±0.20;the Asp(μmol·g -1 ) :3.39±0.12,2.96±0.11, 2.92±0.16, 2.89±0.15 ;the Ca 2+ (μmol·kg -1 ) : 5.84 ±0.11,5.33±0.10,5.28±0.17,5.24±0.12), all being obviously decreased ( P <0 05) compared with those of the chronic gradual spinal cord injury group(Glu 6.32±0.10 μmol·g -1 ; Asp 3.56± 0.13 μmol·g -1 ;Ca 2+ 6.53±0.64 μmol·kg -1 ). They lowered faster 1~7 d, after that, they changed a little. After decompression, the results of open field at 1, 7, 14 and 28 d were 3.53±0.11,3.74±0.12, 3.86± 0.12,3.88±0.10;the results of inclined plane tests were 43.3±2.3,46.2±1.1,48.8±2.8,49.7±2.6. Spinal cord neuronal function had apparently recovered ( P <0 05) compared with that of the chronic gradual spinal cord injury group ( open field 3.40±0.13; inclined plane tests 40.9±1.8), and it had apparently recovered 1~10 d, and after that,there was no apparent change.However, regional water contents of rat spinal cord tissue 1, 7, 14, 28 d after the decompression were 0.7135±0.03,0.7098±0.05,0.7213±0.03,0.6826±0.03,0.7035±0.03, There are no apparent change after the decompression ( P >0 05) compared with those of the chronic gradual spinal cord injury group (0.7135 ±0.03).CONCLUSION : After decompression of spinal cord decompression injury in rats, the contents of excitatory amino acids and calcium ions obviously decrease, which is related with neuronal function recovery of rats.
