柔红霉素对家兔晶体后囊膜细胞的作用
Effect of daunomycin on rabbit lens posterior capsule cells
【摘要】 目的 :观察家兔晶体后囊膜细胞在柔红霉素作用下的组织学及超微结构变化 .方法 :家兔 2 0只随机分为两组 ,分别行白内障囊外摘除人工晶体植入术 .实验组术前于晶体前囊下注入 8mg·L-1柔红霉素 0 .2mL ,对照组同量乳酸林格液 ,作用 5min .术后 7,14和 2 8d处死家兔 ,18只眼球行后囊膜、角膜、虹膜睫状体显微镜观察 .2只行后囊膜透射电镜观察 .结果 :7d时两组后囊晶体上皮细胞增殖 ,14d时实验组后囊膜细胞密度与对照组相比降低 (7.6± 2 .8,12 .8± 4 .8,P =0 .0 39) ,2 8d时实验组后囊膜细胞退变 ,密度进一步降低 (3.5± 0 .9,9.3± 1.3,P =0 .0 0 1) .电镜下对照组细胞结构完整 ;实验组细胞退变 ,核质溶解 .结论 :柔红霉素在 8mg·L-1剂量下 ,采用术前晶体前囊下注入的方法 ,短期内能够抑制家兔晶体后囊膜细胞增殖
【Abstract】 AIM: To evaluate the pathologic features of rabbit lens posterior capsule cells under the effect of daunomycin. METHODS: Twenty rabbits were randomly assigned into two groups: The control group and the experimental group for ECCE (extra capsular cataract extraction)+IOL (intraocular lens) operation. Before lens capsules were truncated, 0.2 mL of daunomycin (8 mg·L -1 ) were injected into anterior capsular in the experimental group; while the same volume of lactic acid Ringers solution was used in the control group. The solution was let to permeate to the ambitus in 5 min. Then the operation was done in routine.18 rabbits were killed 7, 14 and 28 d after the operation, and the lens posterior capsule cells, cornea, iris and ciliary body were observed under the light microscope. 2 rabbits were killed 28 d after the operation, and the posterior capsules were observed under the transmission electron microscope (TEM). RESULTS: More fibrosis and vacuolated cells proliferation of lens epithelial cells could be seen 7, 14 and 28 d after the operation in the control group, While in the experimental group, 7th on the POD (postoperation day) the situation was similar to that of the control group, 14th POD rarefaction fibrosis and vacuolated cells proliferation in posterior capsule cells could be seen, and nuclei degenerated with obviously lowered cellular density(7.6±2.8, 12.8± 4.8, P =0.039); on the 28th POD the nuclei degenerated with low cellular density progressed(3.5±0.9, 9.3±1.3, P =0.001). Under TEM, the cells in the control group were tightly arrayed with clear cellular boundary and structure; while in the experimental group, the cellular deformity and uncear innocellular structure and nuclear cytoplasmic lysis could be noticed. CONCLUSION: Injection of daunomycin (8 mg·L -1 ) into anterior capsular inhibited proliferation of lens posterior capsule cells.
