3种磨损颗粒对体外原代培养人成骨细胞的作用
Effects of 3 kinds of wear particles on primary cultured human osteoblasts
【摘要】 目的 比较 3种不同磨损颗粒对体外原代培养人成骨细胞的作用。方法 PE颗粒、TI 6Al 4V颗粒、Co Cr Mo颗粒加入体外原代培养的人成骨细胞 ,噻唑蓝 (MTT)比色试验检测细胞存活和生长。用酶联免疫检测仪在 490nm波长处测定其吸光度值 ,可间接反映活细胞数量。结果 不同浓度、不同磨损颗粒对原代培养成骨细胞成活无明显影响。结论 不同浓度、不同磨损颗粒对原代培养成骨细胞成活无明显影响 ,表明人工关节松动中磨损颗粒的骨溶解作用是通过其它途径起作用的
【Abstract】 Objective To compare the effects of 3 kinds of wear particles on primary cultured human osteoblasts. Methods After PE, TI 6Al 4V and Co Cr Mo particles were added into primary cultured human osteoblasts, the viability and growth of the osteoblasts were measured by MTT assay. Optical density(OD) at 490 nm was determined by ELISA. Results Wear particles of different kinds and concentrations had no effect on viability and growth of osteoblasts. Conclusion The clinical findings suggest that osteolysis during aseptic loosening is by other route instead of the direct action of wear particles on osteoblasts.
