大鼠创伤性脑损伤后NGF mRNA表达变化及外源性IL-1β对其影响
Changes of NGF mRNA expression and effect of exogenous IL-1β on it in rat after traumatic brain injury
【摘要】 目的 观测大鼠脑损伤后NGFmRNA表达变化及IL 1β对其的影响 ,探讨NGF及IL 1β在脑创伤中的作用机制。方法 在建立流体脑创伤模型的基础上 ,采用RT PCR、分子杂交及免疫细胞化学等技术 ,观测脑创伤后NGFmRNA的表达变化以及外源性IL 1β对其的影响。 结果 NGFmRNA在伤后 12h脑伤灶及其周围即出现表达的增加 ,在脑致伤后2 4h及 3dNGFmRNA的表达明显增加 ,达到峰值 ,其后逐渐降低但表达仍然高于对照组。IL 1β处理组致伤后 3h即出现NGFmRNA表达的增加 ,NGFmRNA的含量明显高于单纯致伤组及对照组 (P <0 .0 1) ,并在伤后 2 4h达到峰值。结论 脑创伤后NGFmRNA表达的增加可能和创伤早期对神经细胞的保护作用和修复机制有关。IL 1βmRNA的表达增加早于脑创伤后NGFmRNA的表达增高。外源性IL 1β能提高脑创伤后NGFmRNA的表达水平。
【Abstract】 Objective To investigate the changes of NGF mRNA expression and the effect of exogenous interleukin 1β(IL 1β) on it in rats after traumatic brain injury(TBI) and to explore the mechanism of NGF and IL 1β in TBI. Methods A brain trauma model of fluid percussion in rats was established. Changes of NGF mRNA expression and effect of exogenous IL 1β on it were observed by RT PCR, molecular hybridization and immunocytochemical techniques. Results NGF mRNA expression in the brain injury site and tissues adjacent to it began to increase at 12 h after trauma and increased markedly at 24 h and reached the peak value on the 3rd. Then it decreased gradually, but still higher than that of the control. NGF mRNA expression increased at 3 h after trauma in IL 1β treatment group and was significantly higher than that in simple trauma and control groups( P <0.01) and peaked at 24 h following brain trauma. Conclusion High expression of NGF mRNA after TBI may be related to the mechanisms of neuron protection and repair. Exogenous IL 1β can increase NGF mRNA expression level following brain trauma.
