新基因RS1在辐射损伤小鼠的组织表达谱研究

【摘要】 目的 探讨新基因RS1在辐射损伤小鼠的组织表达谱情况 ,为RS1的克隆和功能研究提供更多的线索。方法 利用半定量RT PCR的方法研究RS1在辐射损伤小鼠肠上皮、肝、肾、脾脏的表达情况 ,利用原位杂交的方法观察RS1在辐射损伤小鼠肝、肾、脾脏和心肌的表达情况。结果 RS1在辐射损伤小鼠除在肠上皮表达以外 ,在肾、肝、脾等多处表达 ,其中脾脏和肾脏的表达较高。结论 RS1具有广泛的组织表达谱。可以利用肾脏和肝脏来源的 ,RS1表达丰度高的总RNA为模板进行全长cDNA克隆

【Abstract】 Objective To explore the expression profile of RS1 in different radiation injured mouse tissues for the purpose of providing basis for the studies of full length cDNA cloning and functions of RS1. Methods The expressions of RS1 in the intestinal epithelium, liver, kidney and spleen of the mouse exposed to γray were determined by semi quantitative reverse transcription polymerase chain reaction (RT PCR). In situ hybridization was employed to examine the expressions of RS1 in the liver, kidney and spleen of radiation injured mice. Results Expression of RS1 was found in the intestinal epithelium. But there was higher expression in the liver, kidney and spleen of the radiation injured mice. Conclusion The expression profile of RS1 is wide in mouse tissues. The total RNA extracted from the kidney and liver is the optimal sample applied to clone the full length cDNA of RS1.
