纤维连接蛋白在小鼠胚胎干细胞诱导为神经前体细胞中的作用
Effects of fibronectin on the induction of neural precursor cells from murine embryonic stem cells
【摘要】 目的 观察采用ITSF和N2 无血清培养基诱导小鼠胚胎干细胞为神经前体细胞的作用及纤维连接蛋白对神经诱导的影响。方法 小鼠胚胎干细胞的培养和无血清诱导为神经前体细胞 ,NBT BCIP染色 ,Nestin免疫组化染色。结果 细菌培养皿法或悬滴法培养胚体、ITSF或N2 无血清培养基均能将小鼠胚胎干细胞诱导为神经前体细胞。纤维连接蛋白预处理培养皿表面 ,对胚体的贴壁情况影响不大 ,但能使无血清培养后存活的细胞增多。结论 小鼠胚胎干细胞在ITSF和N2 培养基均能分化为神经前体细胞 ,在N2 培养基中加入纤维连接蛋白能使存活的神经前体细胞数量增多
【Abstract】 Objective To observe the effects of ITSF and N 2 serum free medium on the differentiation of neural precursor cells from murine embryonic stem cells and the influence of fibronectin. Methods Cultivation of murine embryonic stem cells and differentiation to neural precursor cells with serum free medium. The embryonic stem cells were stained with NBT/BCIP and the neural precursor cells stained with immunocytochemistry of nestin. Results Murine embryonic stem cells could be induced into neural precursor cells with ITSF or N 2 serum free medium. The number of survival cells in serum free medium increased after the surface of dishes was treated with fibronectin, however, no changes of the number of embryonic bodies were found. Conclusion Murine embryonic stem cells can be differentiated into neural precursor cells in ITSF and N 2 media. In medium N 2 combined with fibronectin, the survival of neural precursor cells can be increased.
