反义c-jun寡核苷酸对大鼠学习记忆和LTP的影响

【摘要】 目的 观察反义c jun寡核苷酸对大鼠学习记忆和长时程增强 (Long termpotentiation ,LTP)的影响。方法 采用脑内微量注射、主动回避反应训练 (AAR)、LTP记录等技术。结果 反义c jun寡核苷酸双侧海马 (5mol L)或脑室注射(8mol L) ,大鼠AAR的习得均明显低于对照组 ;反义c junS ODN使大鼠LTP诱出率明显减少 ,串刺激后PS幅度和fEPSP斜率的增幅降低。结论 反义c jun寡核苷酸显著损害大鼠学习记忆和LTP能力。

【Abstract】 Objective To observe the effect of c jun antisense oligonucleotide on learning, memory and LTP in rats. Methods Techniques such as intrahippocampal microinjection, active avoidance reaction(AAR), long term potentiation(LTP) recording, etc . were used. Results Repeated intrahippocampal(5 mol/L) or intracerebroventricular(8 mol/L) injection of antisense c jun S ODN for three days impaired the learning and memory in active avoidance reaction in young rats. Antisense c jun S ODN injected intrahippocampally (5 mol/L) reduced the probability of the induction of hippocampal LTP in vivo in young rats. The amplitude of population spike (PS) and the slope of field excitatory postsynaptic potential(fEPSP) also decreased following tetanic stimulation. Conclusion c jun antisense oligonucleotide may play an important role in learning, memory and LTP in rats.
