从皂苷糖基水解酶产生菌sp.48g提取基因组DNA
Genomic DNA extracted from sp.48g strain producing saponing-glycosidase
【摘要】 采用试剂盒法、饱和酚抽提法、高盐沉淀法和氯化苄法从皂苷糖基水解酶产生菌sp.48g提取基因组DNA。经比色法和琼脂糖凝胶电泳分析检测。结果用氯化苄法提取的基因组DNA在质量上和得率上优于其他方法。A260/A280为1.7,DNA产量为0.11mg/g湿菌体。达到下部实验要求。
【Abstract】 We extract genomic DNA and isolate it from sp.48g mycelia by the following methods:reagent kit method,liquid frindssphenol extraction, highconcentrationsalt precipitation, and benzyl chloride. The quality of the DNA are identified by agarose gel elcetrophoresis. The purity of DNA is determined by colorimetry. It shows that the quality and yield of DNA samples obtained by benzyl chloride method ( DNA yield is 0.11?mg/g wet mycelium; ratio about A260/A280 is 1.7) are superior to those by other methods, fulfilling the experimental requirements for next step.
