蛋黄磷脂的反相HPLC法分离
The Determination of Yolk Phospholipids by Reversed Phase -High Performance Liquid Chromatography
【摘要】 对北京奥博星生物技术责任有限公司产蛋黄卵磷脂、华东师范大学化工厂(上海)产卵磷脂、北京清华紫光公司产卵磷脂胶囊和自产蛋黄卵磷脂在C18反相柱上进行了反相HPLC法分离,流动相:乙腈-甲醇-水(40:40:20 v/v).检测了所有样品在200~250nm范围内的吸收特性,确定最佳测定波长为205nm.所有磷脂样品均用三氯甲烷溶解.
【Abstract】 A successful method for the determination of yolk phospholipids by reversed phase -high performance liquid chromatography is developed. The samples were separated by RP-HPLC on column HYPERSIL C18 5靘 (250mm?.6mm ID). The best condition for determination is as follows: mobile phase is acetonitrile , methanol , water (40:40:20 v/v) ; flow rate is 1.2mL/min ; column temperature is 30℃; detector is SPD-M10A (SHIMADZU). The absorption of the chloroform (the solvent) and all the samples were determined in the ranges of 200nm to 250nm; and UV 205nm is best.
