离体大鼠脾淋巴细胞在高分压氧下产生羟自由基
Free radical generation by splenic lymphocyte after in vitro exposure to hyperbaric oxygen in rats
【摘要】 目的 :探讨高分压氧暴露时离体脾淋巴细胞是否产生自由基及其产生类型。 方法 :(1)在大鼠脾脏淋巴细胞培养液中分别加入超氧化物歧化酶 (SOD)、过氧化氢酶 (CAT)和甲酸钠 ,置于压力舱内以 4 0 0 k Pa含 1.5 % CO2 氧气培养 4 h后 ,测定细胞经刀豆球蛋白 (Con A)作用后分泌 IL- 2的能力 ;(2 )在大鼠脾脏淋巴细胞悬液内加入自旋捕捉剂四特丁氮酮 (PBN) ,在6 0 0 k Pa含 1% CO2 的氧气下暴露 1h,以电子自旋共振 (ESR)技术检测自由基。 结果 :高分压氧暴露后脾淋巴细胞经 Con A刺激分泌的 IL- 2显著减少 (P<0 .0 1) ,培养液中添加 CAT和甲酸钠后 ,细胞分泌 IL- 2的能力恢复 (P<0 .0 1和 P<0 .0 5 ) ,SOD没有上述作用 ;ESR检测到淋巴细胞在高分压氧下产生自由基 ,其谱型为一组峰强度比约 1∶ 2∶ 2∶ 1的四峰波谱 ,波谱学分析为 HO·加合物的谱线。 结论 :高分压氧体外暴露时 ,大鼠脾淋巴细胞产生自由基 ,HO·可能是最主要的一种
【Abstract】 Objective: To explore which kind of free radical is generated in splenic lymphocyte exposured in vitro to hyperbaric oxygen.Methods: Splenic lymphocytes of adult rats were separated and cultured under 400 kPa O 2 (containing 1.5% CO 2) in a temperature controlled hyperbaric chamber for 4 h.The culture media contained superoxide dismutase (SOD),catalase (CAT) or sodium formate,respectively.The excretion of IL 2 after ConA stimulation was determined with ELISA.Lymphocytes were exposed for 1 h to 600 kPa O 2 (containing 1% CO 2) after adding spin trapping agents N t butyl alpha phenylnitrone (PBN) in the medium and the free radical was determined with electron spin resonance (ESR).Results: After hyperbaric oxygen exposure,the IL 2 excreting ability of splenic lymphocytes was reduced ( P <0.01) and recovered by adding CAT ( P <0.01) and sodium formate ( P <0.05) in the medium,but SOD had no above effect.An ESR spectrum with a 1∶2∶2∶1 quartet pattern and the characteristic of HO· adduct was detected.Conclusion:Splenic lymphocytes can produce reactive oxygen species under hyperbaric oxygen and HO· may be the major free radical generated under this circumstance.
