离子浓度对肽核酸压电基因传感器杂交效应的影响
Effects of ion concentrations on hybridization efficiency with PNA piezoelectric gene sensor
【摘要】 目的 探索不同离子浓度的PBS缓冲液对肽核酸压电基因传感器阵列杂交效应的影响。方法 压电基因传感器阵列表面固定上 1 .5 μM的bis PNA探针后 ,分别观察在 1 0、2 0、5 0、1 0 0mM的PBS缓冲液环境中与相应的靶序列杂交 ,记录频率的下降值及反应所需的时间。结果 钠离子浓度从 1 0mM增加至 2 0mM时 ,杂交导致的频率下降值及平衡时间均增加 (P <0 .0 5 ) ;从 2 0mM增加至 5 0、1 0 0mM时 ,杂交导致的频率下降值各组间无显著差异 ,但杂交平衡时间却呈倍数增长。结论 离子浓度极大地影响了杂交反应的时间 ,低盐离子浓度更有利于提高bis PNA和dsDNA的杂交速率 ,高盐离子浓度则会大大降低bis PNA的结合速率。
【Abstract】 Objective To investigate the effects of ion concentrations on hybridization efficiency with peptide nucleic acid (PNA) piezoelectric gene sensor array.Methods 1.5μM bis PNA probe was firstly immobilized on the surface of gene sensor array.Then it was hybridized with corresponding target sequence when ion concentrations of PBS varied from 10mM to 100mM,and the decrease of frequency and equilibrium time of reaction were recorded and analyzed.Results Both of frequency change and equilibrium time were all increased when ion concentrations varied from 10mM to 20mM( P <0.05).But when it varied from 20mM to 100mM,difference of frequency change was not evident,however,equilibrium time was increased significantly.Conclusion Hybridization time was effected greatly by ion concentrations.Hybridization rate of bis PNA reacting with dsDNA was improved by low ion concentration,while it was reduced significantly by high ion concentrations.
