牦牛生长激素基因的测序和多态性研究
Sequencing and Polymorphism Analysis on Growth Hormone Gene in Yak
【摘要】 采用 PCR产物直接测序法获得牦牛生长激素基因 ( GH基因 ) 891 bp序列。通过 Gen Bank中的 Blastn软件进行序列比较分析表明 ,牦牛 GH基因与普通牛 GH基因的同源性为 98% ,与水牛、山羊、绵羊同源性分别为 96%、 94%、 93% ;NJ法构建的分子系统树显示 ,牦牛与普通牛亲缘关系最近 ,其次为水牛 ,而与山羊、绵羊的亲缘关系较远。此外 ,采用 PCR- RFLP方法研究了 30头牦牛GH基因 891 bp DNA片段 ,未发现 Msp /Hpa 酶切位点多态性 ,表明牦牛 GH基因的多态性比较贫乏。
【Abstract】 In this paper, GH gene from intron 2 to intron 4 were sequenced in Yak. The comparison of the fragment sequence with those of Bos taurus, Bubalus bubalis, Capra hircus, Ovis aries GH gene through blastn of GenBank. The results indicated that the homologies of the nucleotide sequence with Bos taurus were 98%, and with Bubalus bubalis, Capra hircus , Ovis aries were 96%, 94%, 93%, respectively. Molecular phylogenetic tree on ruminant constructed by NJ method ,which showed that molecular tree together with species trees was unanimously. In addtion, it revealed an Msp Ⅰ/Hpa Ⅱ site polymorphism in amplified fragments in GH gene from partial intron 2 to intron 4 of Yak by PCR RFLP. There existed only one genotypes BB in Yak. AA, AB genotpye hadnt been discovered.
