血府逐瘀汤对缺血再灌注损伤大鼠心肌细胞的保护作用研究
Protective Function of Xuefu Zhuyu Tang on Rats’Myocardial Cells with lschemic Reˉperfusion Injury
【摘要】 目的 :观察血府逐瘀汤对缺血再灌注损伤大鼠心肌细胞损伤的保护作用。方法 :利用培养的乳鼠心肌细胞造成缺血再灌注损伤模型 ,观察心肌细胞中超氧化物歧化酶(SOD)的水平来间接推断细胞内活性氧的生成量 ;观察培养液中乳酸脱氢酶(LDH)的量以判断细胞损伤情况 ,并进行DNA琼脂糖凝胶电泳及末端脱氧核苷酸介导的脱氧尿苷三磷酸缺口末端标记法(TUNEL)以判断细胞死亡类型及程度。运用以上指标来判断血府逐瘀汤对心肌细胞缺血再灌注损伤的保护作用。结果 :血府逐瘀汤可显著升高缺血再灌注时SOD水平 ,显著降低LDH水平 ,DNA电泳图谱表明 :血府逐瘀汤可使DNA拖尾基本消失 ,TUNEL结果亦表明血府逐瘀汤可显著减少缺血再灌注所致的心肌细胞凋亡。结论 :血府逐瘀汤有保护缺血再灌注时心肌细胞免于死亡之功效。
【Abstract】 Objective:observe the protective function of Xuefu Zhuyu Tang(XZT)on rats’myocardial cells with ischemic reˉperfusion injury.Method:Make the model of ischemic reˉperfusion injury with the cultured suckling mouse’s myocardial cells,observe the SOD level in the myocardial cells to indirectly induce the production of active oxygen in the cells;observe the LDH content in the culture fluid for the state of cell injury,make DNA agarose gel electrophoresis and dUtriphosphate gap end mark mediated by end deoxyribonucleotide(TUNEL)to define the type and degree of cell death,use the data above to judge the protective function of XZTon myocardial ischemic reˉperfusion injury.Result:XZTcan markedly raise SOD level and lower LDH level at ischemic reˉperfusion.The DNA electrophoresis spectrum shows that XZT can basically disappear DNA tail;the TUNEL also indicates that XZTcan apparently reduce the myocardial cell apoptosis caused by ischemic reˉperfusion injury.Conclusion:XZThas the function to protect myocardial cells from death at ischemic reˉperfusion.
