肿瘤坏死因子在宫颈细胞系表达及其杀伤作用

【摘要】 ①目的 探讨肿瘤坏死因子α(TNFα)mRNA在宫颈细胞系的表达及TNFα对宫颈细胞系的杀伤作用。②方法 采用非放射性核素原位杂交法 ,检测了 3个传代的宫颈细胞系TNFαmRNA的定位表达 ;同时采用LDH释放细胞毒实验 ,观察TNFα对传代的人宫颈癌细胞系的杀伤作用 ,以及人干扰素 (IFNr)对TNFα产生的细胞毒性的协同作用。③结果 TNFαmRNA在SiHa和W 12细胞中有阳性表达 ,NCx细胞无阳性表达。TNFα对宫颈良、恶性肿瘤细胞均有微弱的细胞毒作用 ,而IFNr与TNFα联用则对宫颈癌来源的细胞系具有明显杀伤作用 (F=11.2 5 ,P <0 .0 1) ,对来源于正常、良性肿瘤的细胞系则无明显作用 (F =1.15、1.2 3,P >0 .0 5 )。④结论 宫颈人乳头瘤病毒感染对细胞内TNFα表达有促进作用 ;TNFα对宫颈细胞系具有杀伤作用 ,而且IFNr能促进这种杀伤作用。

【Abstract】 Objective\ To investigate the expression of tumor necrosis factor (TNF) in the cervical cell lines and its cytotoxic effect on cervical cell lines; and observe the synergetic cytotoxicity of interferon (IFNr)and TNFα.\ Methods\ A non radionucide in situ hybridization assay was used to detect the locus expression of TNFα mRNA in three cervix derived cell lines; and a cytotoxicity assay (LDH reiease method ) was applied to detect the cytotoxic effect of TNFα on cervical cell lines and its synergistic action with IFNγ.\ Results\ TNFα mRNA expression was detected in SiHa and W12 cell, there was no TNFα mRNA expression in NCx cells. TNFα had little cytolytic effect to all the cell lines used. However, with the synergistic effect of IFNr, TNFα showed significant cytolytic effect on cervix carcinoma cells ( F=11.25, P <0.01),but no significant effect on normal cells and benign cervix tumor cells were observed ( F=1.15, 1.23, P >0.05). \ Conclusion\ TNFα mRNA expression was promoted when the cervical cells were infected with HPV. TNFα had cytolytic effect on tumor cells and the effect could be enhanced by IFNr.
