利用标记基因检测转Bt基因棉种子纯度的研究
Technique for Testing Seeds Purity of Bt Transgenic Cotton by Using Marker Gene
【摘要】 根据转Bt基因棉"国抗1号"转入的外源基因中整合有GUS基因和NPT 基因,建立了3种种子纯度检测方法:GUS组织化学染色法、NPT 培养基法和NPT 叶片涂抹法,准确率都达到98%以上;在检测转Bt基因棉原始群系"20-1"时与生物测定比较准确率也在98%以上,因此这3种方法可用于转Bt基因棉大规模制种过程中和推广过程中良种纯度的检测,相应方法同样适用于基因组中整合有GUS基因和(或)NPT 基因的其它转Bt基因棉的品种。
【Abstract】 According to GUS gene and NPTⅡgene transformed into Bt transgenic cotton cultivar "Guokang mian No1",three methods for testing the purity of the cotton seeds were established.The results indicated that the accurate rates were up to 98%,and the accurate rates were also up to 98% (compared to the result of bioassay) in testing the original Bt transgenic cotton group "201".The corresponding methods were suitable to every kind of Bt transgenic cotton cultivar with GUS gene or (and) NPTⅡgene.
