一种制作组织芯片的新方法
A Novel Method for Fabrication of Tis sue Microarray
【摘要】 背景与目的:组织芯片(组织微阵列)是继基因芯片和蛋白质芯片之后出现的一种重要的生物芯片技术,是后基因组时代基因组学和蛋白质组学研究的重要方法之一。为了克服现有组织芯片仪器不够先进、操作繁琐、取样点样精度低等缺点,拟设计一种制作组织芯片的新方法。方法:在普通光学显微镜上加装特制的打孔针和取样针,在显微镜载物台上安置特制的石蜡块固定盒,利用显微镜精密的机械控制功能,进行受体石蜡块打孔和供体石蜡组织的取样、点样,并借助显微体视镜进行精确的取样部位观察与定位。结果:简化了组织芯片制作方法,使打孔、定位、取样和点样操作在同一仪器同一操作平台完成,并首次采取一次性使用的打孔针和取样针,提高了取样精度,避免了不同标本之间的组织残留和污染,成功制作了高质量的大肠肿瘤组织芯片。结论:改装普通显微镜用以制作组织芯片方法简便,技术可靠,费用低廉,值得推广。
【Abstract】 BACKGROUND &OBJECTIVE:Tissue chip(tissue microarray,TMA)is one of the most important biochip t echniques,just following the g ene chip and the protein chip,which is on e of the most important functional g e nomics and proteomics research methods in t he post-g enomic era.However,the present TMA technolog y has certain s hortcoming s,such as lack of advanced instruments ,tedious procedure,an d low sampling accuracy,etc.This new method was desig ned to improve the te chnolog y of TMA.METHODS :A common microscope was installed wit h a special holing needle,a sampling needle,and a proper box to fix paraffin blocks on the microscope carrier.With the precise mechanical control of mi croscope,the holing procedure on th e recipient paraffin blocks and sampl ing procedure of core tissue biopsie s were performed with the re-equipped micr oscope.The precise observation and localization of sampling reg ions we re also performed using the same micr oscope equipped with a stereoscope.RESULTS :The new method simplified TMA procedure,and the whole process of h oling ,locating ,and sampling was performed with the same instrument o n the same platform.The sing le-use h oling and sampling needles were first appl ied to maintain hig her accuracy and t o avoid the tissue remains and contaminatio n among different samples.And hig h-qualified tissue chips of colo rectal tumors were produced successfully by the new method.CONCLUSION:Using the re-equipped common micros cope to fabricate tissue microarrays is a si mple,reliable,cost-effective,and well-applicable technique.
