RAPD技术在中药贝母类研究中的应用

【摘要】 目的:为了对贝母类药材的正确应用提供分子水平的资料。方法:采用RAPD技术对12个贝母样品进行了亲缘关系研究。结果:所有样品总DNA均为21 Kb左右。从20个随机引物中筛选出5个扩增稳定且谱带清晰的引物,扩增产物共记录到27条谱带,多态性片段25条,扩增产物大小为450 bp~1904 bp。结论:种内相似性大于种间相似性,安徽贝母和蒲圻贝母的亲缘关系最远,浙贝母和蒲圻贝母的亲缘关系最近。

【Abstract】 Objective:To provide the molecular data for the right application of the Chinese medicine "Beimu".Method:Using the technology of RAPD,we studied the relationship among 12 samples of Beimu.Result:The total genomic DNA of all the samples are about 21 Kb in size.Among 20 primers used,the five primers can reatedly generate a certain specified amplified band type,27 bands were recored from all amplified products and 25 polymorphic fragments were found in it.The size of amplified fragments is between 450 bp and 1904 bp.Conclusion:The similarity within species is higher than those between species.The relationship of Fritillaria anhuiensis and F.puqiensis is the rarest,while F.thunbergii and F.puqiensis is the closest.
