【摘要】 目的:通过测定脑和脾脏中乙酰胆碱酯酶(AchE)、胆碱乙酰化转移酶(ChAT)活性和 M-受体结合容量(Rt)等指标,探讨补肾中药延缓脑功能衰老的机理。方法:用微量酶底物比色法测定 AchE 活性,放射酶分析法测定 ChAT 活性以及用放射性标记配基受体结合方法测定 M 受体结合容量。结果:老年大鼠脑和脾脏中 AchE 活性比青年对照组显著升高,ChAT 活性明显降低,M-受体 Rt值也显著降低。SRBC 免疫后,脑和脾脏中 AchE 活性较未免疫的老年大鼠更高,ChAT 活性降低,表明外来抗原产生的免疫应答可通过增加 AchE 活性及降低 ChAT 活性而降低 Ach 含量。补肾中药可抑制未免疫和 SRBC 免疫后老年大鼠脑和脾脏中 AchE 活性,增加 ChAT 活性,增加 M-受体 Rt 值,改善脑的记忆功能。结论:补肾中药可以通过调节 Ach 的合成和分解酶活性以增加脑和脾脏组织中 Ach 含量,提高 M 受体 Rt 值,从而提高老年大鼠脑记忆功能。更多还原

【Abstract】 Objective:To study the mechanism of Chinese herbal medicine for tonifying Kidney in delaying the senility of brain function.Methods:Acetylcholinesterase(AchE),cholineacetyltransferase(ChAT)activity and M-receptor,total concen- tration(M-Rt)in brain and spleen were determined by enzyme-substract microcolorimetric analysis,radio-enzyme assay and receptor radio-ligand binding assay respectively.Results:AchE activity was increased,CHAT lowered and MR de- creased in brain and spleen of senile rats significantly as compared with those of young rats.By immunizing with SRBC, the changes of AchE and ChAT activity in brain and spleen of senile rats were even more significant,indicating that im- mune response caused by external antigen could reduce content of acetylchotine(Ach)through increasing AchE and lower- ing ChAT activity.Chinese herbal medicine for tonifying Kidney could improve memory function of brain through inhibit- ing AchE activity,increasing ChAT activity and M-Rt of brain and spleen in both unimmune and SRBC immunized senile rats.Conclusion:Chinese herbal medicine for tonifying Kidney could increase content of Ach and M-Rt by regulating com- position and resolution of Ach in brain and spleen so as to improve the memory function of brain in senile rats.更多还原
